What it is

Tesamorelin is a synthetic 44-amino-acid GHRH analogue modified for stability. FDA-approved (2010) for reduction of visceral adipose tissue in HIV-associated lipodystrophy.

Evidence snapshot

Category: Growth Hormone. Status: Human Trials. See the references section below for the underlying primary sources.

Mechanism

Binds the GHRH receptor, stimulating endogenous GH release and downstream IGF-1 elevation. Pulsatile GH release more closely mimics physiological patterns than exogenous GH.

Key research findings

Phase 3 trials demonstrated significant reduction in visceral adipose tissue with tesamorelin treatment in HIV-associated lipodystrophy.

Limits of the evidence

Limited indication (HIV-associated lipodystrophy). IGF-1 elevations can exceed physiological range; monitoring recommended. Off-label use in other indications lacks rigorous trial support.
