Retatrutide: Triple GIP/GLP-1/Glucagon Agonist

A triple-receptor agonist (GIP/GLP-1/glucagon) currently in Phase 2/3 trials for obesity and metabolic disease.

What it is

Retatrutide is a synthetic peptide combining GIP, GLP-1, and glucagon receptor agonism in a single molecule. Developed by Eli Lilly.

Evidence snapshot

Category: Metabolic. Status: Human Trials. See the references section below for the underlying primary sources.

Mechanism

Activates GIP and GLP-1 receptors (reducing appetite) and the glucagon receptor (increasing energy expenditure). The combination is hypothesised to produce greater weight loss than dual-agonists.

Key research findings

Phase 2 trial data in obesity showed dose-dependent weight loss up to ~24% over 48 weeks — among the largest reported in any obesity pharmacotherapy development program.

Limits of the evidence

Phase 3 data not yet published. Long-term safety of chronic glucagon agonism requires longer follow-up. Adverse event profile includes GI effects.
