Melanocortin
Compound profile

Melanotan II: Non-Selective MCR Agonist

A non-selective melanocortin receptor agonist with affinity for MC3R, MC4R, and MC5R in addition to MC1R.

What it is

Melanotan II is a synthetic cyclic heptapeptide developed alongside Melanotan I. Activates multiple melanocortin receptors, unlike the more selective Melanotan I.

Evidence snapshot

Category: Melanocortin. Status: Preclinical. See the references section below for the underlying primary sources.

Mechanism

Activates MC1R (pigmentation), MC3R/MC4R (sexual arousal, appetite), and MC5R (exocrine gland function). The MC3R/MC4R activity produces the sexual arousal effects.

Key research findings

Pigmentation effects are well-documented in off-label use. Sexual arousal effects have been reported in small studies.

Limits of the evidence

Off-label use is widespread. Not approved for any human use. Adverse effects include nausea, flushing, and potential for melanoma promotion in predisposed individuals. WADA classifies as prohibited.
