Hexarelin: Synthetic Hexapeptide GHSR Agonist

A synthetic hexapeptide ghrelin mimetic studied for GH release, with characteristic tachyphylaxis on repeated dosing.

What it is

Hexarelin is a synthetic hexapeptide GHS developed in the 1990s. Strong agonist at GHSR-1a, with secondary activity reported at the CD36 scavenger receptor.

Evidence snapshot

Category: Growth Hormone. Status: Preclinical. See the references section below for the underlying primary sources.

Mechanism

Activates GHSR-1a to produce GH release. The CD36 activity produces cardioprotective signalling in some preclinical models. Repeated dosing produces rapid tachyphylaxis.

Key research findings

Produces robust GH release in rodents and humans, with greater peak than GHRP-6. Cardioprotective effects in ischemic and pressure-overload heart failure models.

Limits of the evidence

Tachyphylaxis limits therapeutic utility. Most published clinical work was small Phase 1 pharmacokinetic studies; clinical efficacy was never established.
