5-Amino-1MQ: NNMT Inhibitor

5-Amino-1MQ is a small-molecule inhibitor of NNMT studied for effects on metabolic rate, NAD+ levels, and adipose tissue browning.

What it is

5-Amino-1MQ is a synthetic small molecule that inhibits NNMT (nicotinamide N-methyltransferase), an enzyme involved in nicotinamide metabolism.

Evidence snapshot

Category: Metabolic. Status: Preclinical. See the references section below for the underlying primary sources.

Mechanism

Inhibits NNMT in adipose tissue, which alters the methylated-nicotinamide/NAD+ balance and supports adipose tissue browning, increased energy expenditure, and reduced fat mass in preclinical models.

Key research findings

In mouse models of diet-induced obesity, 5-Amino-1MQ reduces fat mass, increases lean mass, and improves metabolic parameters.

Limits of the evidence

Preclinical only. Off-label use widespread despite no human safety data. Pharmacological profile in humans is not established.
